The Impact of mRNA Degradation on Transcriptome Analysis: A Rapid Algorithm for Calculating a Novel Degradation Index using Sequencing Data

Zixuan Xie, Yanghui Sheng, Wubin Qian, Sheng Guo

RNA degradation can significantly compromise RNA-seq data integrity, complicating accurate transcriptomic analysis. Assessing post-RNA-seq data quality is often challenging due to the slow speed of current algorithms. Commonly used indices, such as the Ribosomal Integrity Number (RIN), tend to be biased and offer low resolution.

While the Transcript Integrity Number (TIN) is more accurate, it is computationally intensive. This study introduces a novel index, the RNA Degradation Index (RDI), designed to provide a faster and more efficient assessment of RNA degradation.

  • How RDI offers faster, more accurate RNA-seq data quality assessment than traditional methods.

  • How RDI reduces analysis time from 200 minutes to 8 minutes, maintaining high accuracy.

  • How RDI helps detect RNA degradation, preventing biased transcriptomic data.
